| Description | This ligand binds to Fms-Like Tyrosine Kinase-3 (FLT-3).Human FLT3 ligand also stimulates the proliferation of cells expressing murine FLT-3 receptors. The effects of FLT-3 ligand are synergized by coexpression of G-CSF , GM-CSF , M-CSF , IL3 and SCF . FLT-3 ligand promotes long-term expansion and differentiation of human pro-B-cells in the presence of IL7 or in combination of IL7 and IL3 . Recombinant Human Flt3-Ligand produced in Sf9 insect cells is a glycosylated, polypeptide chain containing 155 amino acids and having migrating on SDS-PAGE at 19kDa. Recombinant Human Flt3-Ligand is purified by proprietary chromatographic techniques. Amino acid sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Thr-Gln-Asp-Cys-Ser. Dimers and aggregates: ≤ 1% as determined by silver stained SDS-PAGE gel analysis. Biological Activity: Recombinant Flt3-Ligand is fully biologically active when compared to standard. The ED50, calculated by the dose-dependent stimulation of the proliferation of human MUTZ-2cells is <3.0ng/ml. Endotoxin: ≤ 0.1ng/ug (IEU/ug) of Recombinant Human Flt3-Ligand. Protein content: Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280nm using the absorbency value of 1.42 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). 2. Analysis by RP-HPLC, using a standard of Recombinant Flt3-Ligand as a Reference Standard. Storage and Stability: Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile dH2O or buffer of choice and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | ||
