Description Flt3 Ligand (flk2 ligand), is a growth factor that regulates proliferation of early hematopoietic cells. Flt3 Ligand binds to cells expressing the tryosine kinase receptor Flt3. Flt3 Ligand, by itself does not stimulate proliferation of early hematopoietic cells, but synergizes with other CSFs and interleukins to induce growth and differentiation. Unlike SCF, Flt 3 Ligand exerts no activity on mast cells. Multiple isoforms of Flt 3 Ligand have been identified. The predominant biologically active form is anchored to the cell surface as the extracellular domain of a transmembrane glycoprotein (209aa). The membrane-bound isoform can be proteolytically cleaved to generate a biologically active soluble isoform. Recombinant human Flt3 Ligand is a soluble 17.6kD protein consisting of 155 amino acid residues comprising the extracellular domain of the transmembrane Flt3 Ligand protein. Source: Recombinant protein corresponding to human Flt-3 Ligand, expressed in E. coli. Biological Activity: Determined by its ability to stimulate the proliferation of human AML5 cells. The expected ED50 is <1.0ng/ml, corresponding to a specific activity of >1x10e6units/mg. Endotoxin: <0.1ng/ug of protein (<1EU/ug) AA Sequence: TQDCSFQHSP ISSDFAVKIR ELSDYLLQDY PVTVASNLQD EELCGGLWRL VLAQRWMERL KTVAGSKMQG LLERVNTEIH FVTKCAFQPP PSCLRFVQTN ISRLLQETSE QLVALKPWIT RQNFSRCLEL QCQPDSSTLP PPWSPRPLEA TAPTA Protein Content: Verified by UV Spectroscopy and/or SDS-PAGE gel. Storage and Stability: Lyophilized powder may be stored at -20°C. Reconstitute with H2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
