Description Recombinant corresponding to aa2-end of human UBA6 (UBE1L2) with N-terminal Flag tag expressed in a Baculovirus infected Sf9 cell expression system (NM_018227). Applications: Suitable for use in conjunction with E2 and E3 for the study of enzyme kinetics, screening inhibitors and selectivity profiling. Other applications not tested. Recommended Dilutions: Assay Conditions: UBA6 (100ng), UBCH7 (2800ng) and Flag-NEDD4 (800ng) were incubated in a 50ul reaction mix containing 50mM Tris, pH 7.4, 4mM ATP, 2mM DTT, 5mM magnesium chloride and 0.5ug biotin-Ub at 37°C for 30 minutes, followed by incubations with anti-FLAG antibody, anti-mouse HRP and reagents for chemiluminescence detection. Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
