| Description | Frizzled5 is an ~62kD seventransmembrane (7TM) glycoprotein that is one of at least ten Frizzled family Wnt receptors (1 3). Frizzled proteins are thought to be G proteincoupled (1). Wnt engagement of Frizzled, with low density lipoprotein receptorrelated proteins LRP5 or LRP6 acting as coreceptors, stabilizes B-catenin and promotes gene transcription that is important in development and tissue maintenance (1). LRP independent or “noncanonical” Frizzled signaling pathways also exist (1). Human Frizzled5 cDNA encodes 585 amino acids (aa) including a 26 aa signal peptide, a 212 aa extracellular domain (ECD), the 7TM region (aa 213 521), and a Cterminal cytoplasmic domain with a PDZ binding motif (aa 522 585) (2, 5). The ECD includes a cysteinerich region (CRD, aa 29 156) that binds Wnts and is highly conserved among Frizzleds (1, 2, 4). Human Frizzled5 shares 95%, 95% and 96% aa identity with mouse, rat and canine Frizzled5, respectively, within the Nterminal extracellular portion. It also shares 82% aa identity with human Frizzled8. Frizzled5 is expressed in embryonic tissues (telencephalon, pituitary, thalamus, hypothalamus, eye, liver, spleen, lung and kidney), in the adult retina, colon and pancreatic islets, some cancer cell lines, human embryonic stem cells and in some monocyte and lymphocyte populations (2 9). It is required for ongoing survival of neurons in the parafascicular nucleus of the mouse thalamus (5). Null mutations show a role for Frizzled5 in maintenance of yolk sac and placental vasculature (6). Deletion specifically in the eye results in persistence of vitreous vasculature that normally regresses during late development (7, 10). In macrophages, Frizzled5 participates in a CaMKII signaling pathway that promotes inflammation (8, 9). Wnts implicated as ligands for Frizzled5 include Wnt5a, Wnt9b, Wnt10b, Wnt2b, and Wnt 7a (5, 6, 11, 12). Source: Recombinant corresponding to human, Frizzled5 Fc Chimera, Nterminus. Molecular Weight: ~50.1kD Biological Activity: Measured by its binding ability in a functional ELISA. In a 100ul reaction mixture containing biotinylated rmWnt5a at 100 ng/ml and rhFrizzled5/Fc Chimera dilutions, the concentration of recombinant, human Frizzled5/Fc Chimera that produces 50% of the maximal binding response is found to be approximately 520ng/ml. Optimal concentrations should be determined by each laboratory for each application. Endotoxin Level: <1EU/ug (LAL method) Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile buffer. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C.
