| Description | Frizzled-9 (Fzd9, previously called Fzd3) belongs to the Frizzled family of seven-transmembrane glycoprotein receptors, which are signaling receptors for the family of secreted Wnt ligands (1-3). Mouse Frizzled-9 cDNA encodes 592 amino acids (aa) including a 23aa signal peptide, a 207aa extracellular domain (ECD), the 7TM region (aa231-530), and a C-terminal cytoplasmic domain with a PDZ binding motif. The ECD includes a cysteine-rich region (CRD, aa35-156) that binds Wnts and is highly conserved among Frizzleds. Within aa23-186, mouse Frizzled9 shares 100%, 97%, 97% and 96% aa identity with rat, human, canine and bovine Frizzled9, respectively. The human Frizzled-9 gene is within a large region that is deleted in Williams-Beuren syndrome, a developmental disorder affecting personality and cognition (2). Frizzled-9 mRNA is expressed in the mouse central nervous system and myotomes during development, and persists in the hippocampus throughout life (2-4). Adults also show expression in heart, testis, and skeletal muscle (2-4). Cellular expression is detected in proliferating neural precursors, B lymphocyte progenitors, and differentiating osteoblasts (46). Defects in learning, B lymphocyte development, and bone mineralization are observed in mice deleted for Frizzled-9 (4-6). Wnt-2-stimulated signaling through the canonical pathway using LRP5 as a coreceptor is reported, resulting in beta-catenin stabilization and gene transcription (7). However, the canonical pathway is not involved in bone defects of Frizzled-9-/-mice (6). Signaling through the JNK (noncanonical) pathway is shown when Frizzled9-expressing human non-small cell lung cancer cell lines are engaged by Wnt-7a, promoting activation of PPARgamma and expression of cadherin and Sprouty, and reversing transformation (8-10).
