Description | Preparation of HSV-2 antigens: HSV-2 is propagated in mammalian cells using a protocol optimised for maximum particle yield. At the appropriate time, infected cells are washed in saline then disrupted to release virus particles by freeze thaw and sonication. Purification and presentation: PBS lysate: Virus particles released from cells are partially purified by low speed centrifugation to remove cell debris. HSV-2 particles are then purified by sucrose density gradients. This process also includes a DNase step to remove cellular DNA and helps to achieve a high purity. Source: HSV-2, strain G, from a crude lysate. Applications: Suitable for use in ELISA. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Inactivation: 0.1% Triton X-100. Tested by passage on permissive cells for 5 days yielding no CPE. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: No test guarantees a product to be non-infectious. All materials should be handled as if potentially infectious. Generally accepted laboratory practices appropriate for infectious materials should be employed when handling this product. |
