Description | The Pertussis Toxin Mutant contains a modified sequence encoding the enzyme subunit. Virulence of this pertussis mutant is greatly reduced relative to that found with the wild type. The pertussis mutant protein is isolated from the TY-178 strain of Bordetella bronchiseptica which contains a genetically modified sequence encoding the S1 sub unit. Site-directed mutagenesis was used to replace amino acid residues arginine-9 with lysine (R9K) and glutamic acid-129 with alanine (E129A). Genetically inactivated toxin has reduced activity relative to the native toxin and may be used as an antigen or as a carrier to promote an immune response. Source: Pertussis Toxin, Mutant, strain TY-178 from Bordetella bronchiseptica Biological Activity: The lowest concentration of toxin at which a positive response (clustered growth pattern) was obtained was 3.3ng/ml. The adenylate cyclase activity of this lot, in the presence of 1umolar calmodulin, is 1picomole cAMP/min/ug Pertussis Toxin Mutant when assayed by the method of Wolff et al. Endotoxin: ~20EU/mg (Kinetic Chromagenic LAL assay) Storage and Stability: Lyophilized powder may be stored at 4°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at 4°C. Reconstituted product is stable for 6 months at 4°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
