Description | There are seven immunologically distinct botulinum neurotoxins, designated types A-G. Each neurotoxin toxin cleaves one of two target proteins critical for synaptic vesicle fusion to the plasma membrane and neurotransmitter release, either SNAP-25 or Synaptobrevin. The latter is also called VAMP-2. Types A, C and E specifically bind and selectively cleave SNAP-25 while types B, D, F and G cleave Synaptobrevin. Type C also cleaves Syntaxin. The enzymatic activity of these toxins can also be measured using the appropriate native substrate and monitoring the generation of the cleaved product on SDS-PAGE gel electrophoresis. Source: Recombinant protein substrate corresponding to 214aa from C. botulinum SNAP-25, neurotoxins Type A and E, expressed in E. coli. Molecular Weight: ~24.12kD Activity: SNAP-25 can be used as a substrate to assess the activity of Botulinum Neurotoxin Type A, Botulinum Neurotoxin Type A light chain (LcA) and Botulinum Neurotoxin Type E light chain (LcE). A 100uI reaction of 5uM SNAP-25 (12ug) was completely cleaved within one hour at 37°C in 20mM Tris-HCI, pH 8.0, 50mM NaCI using LcA or LcE. Detection of the cleavage of SNAP-25 by Light Chain E is readily apparent by running a sample of the digestion on a coomassie stained polyacrylamide gel. The cleavage of SNAP-25 by Type A Botulinum Neurotoxin or light chain is not detectable on a gel, due to the small cleavage product, but can be visualized by performing a Western Blot with primary antibodies specific to Botulinum Type A cleaved SNAP-25 (Research and Diagnostic Antibodies, Inc.). Storage and Stability: Lyophilized powder may be stored at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
