Description MMP-3 (matrix metalloproteinase 3), otherwise known as stromelysin-1, is a member of a family of secreted zinc-dependent endoproteases, which play a role in the degradation of extracellular matrix (ECM) components, including both fibrillar and non-fibrillar collagens, laminin, basement membrane glycoprotein and fibronectin. MMP-3 can degrade gelatins type I, III, IV, and V, type III, IV, IX and X collagens, as well as fibronectin, laminin, and cartilage proteoglycans. MMP-3 is implicated in wound repair, and tumor initiation, and studies have shown high expression of MMP-3 by monocytes, and that MMP-3 is also expressed by microglial cells, chondrocytes, and by fibroblast-like synoviocytes in patients with rheumatoid arthritis (RA) and osteoarthritis. Source: Recombinant protein corresponding to human MMP-3 expressed in E. coli. Molecular Weight: ~42.8kD (378aa) containing the entire catalytic N-terminal domain and the C-terminal domain. Activity: MMP-3 activity was measured by its ability to cleave a chromogenic peptide MMP-3 substrate at RT. At a MMP-3 concentration of ~2.5ug/ml, 50% cleavage was achieved at an incubation time of ~75 mins. Applications: Suitable for use in Functional Assays. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Endotoxin Level: <1EU/ug Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
