| Description | MMP-2 (matrix metalloproteinase 2), otherwise known as gelatinase A, is a member of a family of secreted zinc-dependent endoproteases, which play a role in the degradation of extracellular matrix (ECM) components, including both fibrillar and non-fibrillar collagens, laminin, basement membrane glycoprotein and fibronectin. MMP-2 specifically cleaves gelatin type I, and types IV, V, VII and X collagens, and is also involved in tissue repair, inflammation, tumour invasion, vasculature angiogenesis, and the rupture of atherosclerotic plaques. Defects in the MMP2 gene are responsible for the autosomal recessive osteolysis disorder known as Torg-Winchester syndrome (TWS), or multicentric osteolysis nodulosis and arthropathy (MONA). Source: Recombinant protein corresponding to human MMP-2 expressed in E. coli. Molecular Weight: ~62kD (552aa) containing the entire catalytic N-terminal domain and the C-terminal domain. Activity: MMP-2 activity was measured by its ability to cleave a chromogenic peptide MMP-2 substrate at RT. At an MMP-2 concentration of ~2.5ug/ml, 50% cleavage was achieved at an incubation time of ~25 mins. Applications: Suitable for use in Functional Assays. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Endotoxin Level: <1EU/ug Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | ||
