Description Prepared by the C-terminal derivatisation of ubiquitin with 7-amino-4-methylcoumarin. Acts as a useful and sensitive fluorogenic substrate for a wide range of deubiquitinylating enzymes (DUBs), including ubiquitin C-terminal hydrolases (UCHs) and ubiquitin specific proteases (USPs). Ubiquitin-AMC has been shown to be a sensitive substrate for UCH-L3 (Km=0.039uM) and for Isopeptidase-T (Km=0.17-1.4uM), as well as for studying deubiquitinylating activity where detection sensitivity or continuous monitoring of activity is essential. Sequence: C-terminal derivatisation of ubiquitin with 7-amino-4-methylcoumarin Emission: 460nm Excitement: 380nm Molecular Weight: ~8.6kD Substrate Concentration: 0.01-1uM Enzyme Concentration: 10-100pM (UCH-L3); 10-100nM (Isopeptidase-T) Applications: Suitable for use in Deubiquitinylating Enzyme Activity. Other applications not tested. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
