Description CDNF is a secreted neurotrophic factor that is expressed in brain, neuronal and certain non-neuronal tissues. It has been shown to promote survival, growth and function of dopamine specific neurons. CDNF and its structural homolog MANF, each contain an N-terminal saposin-like lipid binding domain, and a carboxyl-terminal domain, which is not homologous to previously characterized protein structures. CDNF and MANF can prevent 6-OHDA induced degeneration of dopaminergic neurons by triggering survival pathways in a rat experimental model of Parkinson disease. Source: Recombinant protein corresponding to 162aa including 8 cysteine residues from human CDNF, expressed in E. coli. Molecular Weight: ~18.5kD Biological Activity: Determined by its ability to stimulate the proliferation of rat C6 cells. The expected ED50 for this effect is 15-25ug/ml. Endotoxin: <0.1ng/ug of protein (<1EU/ug) AA Sequence: MQEAGGRPGA DCEVCKEFLN RFYKSLIDRG VNFSLDTIEK ELISFCLDTK GKENRLCYYL GATKDAATKI LSEVTRPMSV HMPAMKICEK LKKLDSQICE LKYEKTLDLA SVDLRKMRVA ELKQILHSWG EECRACAEKT DYVNLIQELA PKYAATHPKT EL Storage and Stability: Lyophilized powder may be stored at -20°C. Reconstitute with sterile buffer or ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
