Form Supplied as a lyophilized powder. Reconstitute with 15mM sterile HCl (0.5ml/1mg vial or 5ml/10mg vial) to the vial. After the protein is completely dissolved, add 7.5mM sterile NaOH (0.3ml/1mg vial or 3ml/10mg vial) to bring the pH to approximately 5.2.

Description Leptin (Anti-Obesity Protein) is the protein product of the ob (obese) gene in mice and seems to be involved in appetite control. The ob gene is expressed in adipose tissue and is thought to regulate the body’s fat stores. Mice with the ob/ob genotype also develop a form of diabetes similar to type II (non-insulin dependent) diabetes. Human Leptin is a 16kD protein containing 146 amino acid residues. Source: Recombinant, human Leptin expressed in E. coli. Biological Activity: Human Leptin has been shown to be biologically active in two different mouse obesity models, ob/ob and NZO. Both strains of mice were treated via intraperitoneal injection once daily at a dose of 5ug Leptin/gm of body weight for 7 days. Significant effects on body weight, food consumption, and plasma glucose levels were observed to saline- treated controls. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile HCl. After the protein is completely dissolved, add sterile NaOH to bring the pH to approximately 5.2. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
