| Description | Lipopolysaccharide Binding Protein (LBP, LPSBP) is an ~60kD acute phase protein produced by hepatocytes. It strongly binds to LPS and has been shown to play an important role in the handling of LPS by the host. A number of functions of LBP have been reported. First, LBP transfers LPS to the LPS receptor CD14 on mononuclear phagocytes, leading to an 100 to 1000-fold increased sensitivity of the cells to LPS. Furthermore, LBP can enhance the response of CD14 negative cells by acceleration of LPS binding to soluble CD14, a complex that stimulates these cells. Next, LBP transfers LPS into lipoproteins which neutralizes its biological potency. LBP was demonstrated to protect mice from septic shock caused by LPS or gram negative bacteria. The LBP-peptide, aa86-99, binds lipid A and neutralizes LPS in vivo and in vitro. Source: Purified LBP-peptide (aa86-99) from human LBP protein. Applications: Suitable for use in ELISA and Neutralization. Other applications not tested. Recommended Dilution: Neutraliztion (in vivo, in vitro): Dilutions should be determinined according to the amount of protein to be inactivated. Note: Further dilutions should be made by using protein stabilized PBS, pH 7. Never use fresh serum or plasma. Be Aware: Endotoxin will bind LBP-peptide. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile, endotoxin free PBS. Aliquot to avoid repeated freezing and thawing and store at -70°C. The stock solution should never be thawed more than once. Aliquots are stable for 6 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in protein stabilized PBS, pH 7. | ||
