| Description | Control peptide for C5700-04A and C5700-04B. The enzyme citrate synthase (E.C. 2.3.3.1 [previously 4.1.3.7]) is a pace-maker enzyme, as it controls the first committed step of the Krebs cycle, also called the citric acid cycle. Citrate synthase is localized within cells in the mitochondrial matrix, but is encoded by nuclear DNA rather than mitochondrial. It is synthesized using cytoplasmic ribosomes, then transported into the mitochondrial matrix. Citrate synthase is commonly used as a quantitative enzyme marker for the presence of intact mitochondria. Citrate synthase catalyzes the condensation reaction of the two-carbon acetate residue from acetyl coenzyme A and a molecule of four-carbon oxaloacetate to form the sixcarbon citrate. Oxaloacetate will be regenerated after the completion of one round of the Krebs Cycle. Human CS (chromosome 12p11-qter, 466aa precursor, mature chain 28-466aa) encodes a 466aa protein with 95% homology to its pig homolog. CS enzyme, a homodimer, is located at the Mitochondrion matrix. Citrate synthase is found in nearly all cells capable of oxidative metabolism. Recommended Dilution: ELISA: 1ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile buffer or ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | ||
