Description Hinge region peptide amino acids 213-226 of human RAR gamma [lglwdkfselatkc] (linear). Corresponding antibody R1691-10 is specific for Retinoic acid receptor, gamma isotype. Recognizes both isoforms of RAR gamma: RAR gamma-1 and RAR gamma-2. Species crossreactivity: Human and rat RAR gamma. Retinoids can reverse various premalignant lesions as well as prevent some second primary tumors. The effects of retinoids are regulated by retinoic acid receptors (RAR) and retinoid X receptors, which act as ligand-activated transcription factors. Ligand-dependent transcriptional activation of RARs is a multistep process that ends with the formation of a multimeric co-activator complex on regulated promoters (1,2). Retinoic acid receptor gamma has been shown to be the most important retinoid receptor for regulation of retinoic acid turnover rate and retinoic acid induced growth inhibition (1). While RAR mRNA levels may be useful biomarkers for various diseases, RAR gamma expression is decreased in Barrettís tissues compared with normal tissue (3). Applications: Suitable for use in ELISA and Western Blotting. Other applications not tested. Recommended Dilution: Western blot: 1:1000. TBST (20 mM Tris/HCl (pH 7.6), 137mM NaCl, 1% Tween-20) 5% nonfat milk, 0.05% sodium azide. A ~48kD band can be seen using rat brain hippocampal homogenate. The investigator should determine the optimal working dilution for a specific application. Storage and Stability: Lyophilized powder may be stored at 4°C for short-term only. Stable for 12 months at -20°C. Reconstitute to nominal volume (see reconstitution instructions for peptides) and store at -20°C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
