Description Retinoblastoma (RB) is an embryonic malignant neoplasm of retinal origin. It almost always presents in early childhood and is often bilateral. Spontaneous regression ('cure') occurs in some cases. The retinoblastoma gene RB was the first tumor suppressor gene cloned, and is a negative regulator of the cell cycle through its ability to bind the transcription factor E2F (MIM 189971) and repress transcription of genes required for S phase (Hanahan and Weinberg, 2000 [PubMed 10647931]).[supplied by OMIM] Source: Recombinant fusion protein containing the C-terminal fragment (residues 773–928) of human retinoblastoma protein (Rb) with an N-terminal His6-tag, expressed in E. coli. Application: Suitable for use in Kinase Assay. Other applications not tested. Recommended Dilution: Kinase Assay: Rb is an effective substrate for a number of serine/threonine kinases. Tested using cdk2/cyclin A, active (C2571-95A) to phosphorylate 10ug Rb. Storage and Stability: For long-term storage, aliquot and store at -20°C. Aliquots are stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
