| Description | STAM-1 (Signal Transducing Adapter Molecule 1) is involved in multiple signal transduction pathways through interactions mediated by ITAM, SH3, UIM, and VHS domains located within the protein. Roles in growth factor receptor down-regulation, T-cell development, and induction of DNA synthesis following IL-2 stimulation have been identified. This full-length recombinant protein has no intrinsic deubiquitinase activity, but is useful in stimulating the in vitro activity of the JAMM-class deubiquitinase AMSH. Source: Recombinant protein corresponding to human STAM-1. Molecular Weight: ~59kD Biological Activity: Binds AMSH protein with Kd = ~7uM. In in vitro assays containing 200 nM AMSH, a ~10-fold increase in the rate of hydrolysis of K63-linked diubiquitin FRET substrate was achieved upon the addition of STAM. Half-maximal increase occurred at 0.4uM STAM. Reaction conditions for stimulation of AMSH will need to be optimized for each specific application Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
