Description Human sirtuins function as intracellular regulatory proteins. Sirtuin 2 is a member of the class III histone deacetylases (HDACs) and has been implicated in many cellular processes that include histone deacetylation, gene silencing, chromosomal stability, and aging. The enzymatic activity of class III HDACs is nicotinamide adenine dinucleotide (0.1mM NAD+) dependent and insensitive to HDAC inhibitor trichostatin A. Source: Recombinant protein corresponding to human Sirtuin 2, expressed in E.coli. Molecular Weight: ~43kD Biological Activity: Activity of SIRT2 was tested using the substrate Arg-His-Lys-Lys(Ac)-AFC, which is a fluorogenic, acetylated peptide based on residues 379-382 of p53. Unit Definition: One unit of the recombinant SIRT2 is defined as the amount of enzyme that deacetylates 1pmol of substrate Arg-His-Lys-Lys(Ac)-AFC/minute at 37°C in a reaction solution containing 50mM Tris, pH 7.5, 137mM NaCl, 2.7mM KCl, 1mM MgCl2, 0.1mM NAD+, 1mg/ml BSA. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -70°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
