| Description | Control peptide for C7900-70 (antiserum) and C7900-71 (affinity purified antibody). Synthetic peptideconsisting of 17aa sequence from the mid-region of human cbfa1. No significant homology is seen with CBFA2/PEBP2aB/AML1 and CBFA3/PBEP2aC/AML2 or any runt family of proteins. Species Sequence Homology: mouse cbfa1/osf2: 100%. Core biding factors (CBF) are a family of heterodimeric transcription factors comprised of two subunits: A DNA binding a subunit and non-DNA binding b subunit. CBF alpha is encoded by three distinct genes (CBFA1/PEBP2aA/AML3, CBFA2/PEBP2aB/AML1, and CBFA3/PBEP2aC/AML2), and a b subunit encoded by CBFB/PEBP2b. The CBFa subunits share a conserved 128aa runt domain that is homologous to the Drosophila pair-rule gene runt that plays a role in the formation of the segmented body pattern, sex determination and development of the nervous system. The consensus DNA-binding sequence is PuACCPuCA, originally identified as an enhancer in polyoma virus and murine leukemia virus. The null mutation of cbfa2 and cbfb in mice were embryonic lethal. A disruption in cbfa1 gene product blocked osteoclast development from mesenchyme with no ossification. Mutations or deletion in cbfa1 have been shown to be associated with CCD, a human skeletal disorder named cleidocranial dysplasia. Cbfa1 is (alternatively spliced form 507 and 522aa in human and 528aa or 59aa in mouse) is expressed in bone, thymus and testis but not in brain, lung, heart, spleen, liver or kidney. Cbfa1 is an essential transcription factor in osteoblast differentiation. Applications: Suitable for use in ELISA and Antibody Blocking. Not suitable for use in Western Blot due to low molecular weight. Other applications not tested. Recommended Dilution: Antibody Blocking: 5-10ug per 1ul C7900-70 (antiserum) or per 1ug C7900-71 (affinity purified antibody). ELISA: Coat with 1ug of control peptide (C7900-72) per well. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | ||
