Description Peptide is used to block TCF1 (C63D9) Rabbit mAb reactivity in immunohistochemistry protocols. LEF1 and TCF are members of the high mobility group (HMG) DNA binding protein family of transcription factors which consists of the following: Lymphoid enhancer factor 1 (LEF1), T Cell Factor 1 (TCF1), TCF3 and TCF4 (1). LEF1 and TCF1 were originally identified as important factors regulating early lymphoid development (2) and act downstream in Wnt signaling. LEF1/TCF bind to Wnt response elements to provide a docking site for β-catenin, which translocates to the nucleus to promote the transcription of target genes upon activation of Wnt signaling (3). LEF1/TCF proteins are dynamically expressed during development and aberrant activation of the Wnt signaling pathway is involved in many types of cancers including colon cancer (4,5). Quality Control: The quality of the peptide was evaluated by reversed-phase HPLC and by mass spectrometry. The peptide blocks TCF1 (C63D9) Rabbit mAb by immunohistochemistry. Directions for Use: For immunohistochemistry, add twice the volume of peptide as volume of antibody used in 100ul total volume. Incubate for a minimum of 30 minutes prior to adding the entire volume to the slide. Recommended antibody dilutions can be found on the relevant product data sheet. Storage and Stability: For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
